The stage-specific expression of Rab3 mRNA and protein during larval growth and adult maturation in Drosophila melanogaster was studied with a quantitative in situ hybridization technique and an immunohistochemical technique. In situ hybridization revealed the presence of Rab3 mRNA in the epidermis during late larval stages and throughout adult life. Rab3 protein was detected only during adult life in imaginal discs, brain, salivary glands, and Malpighian tubules. Developmental stages of these organs coincided with alterations in the Rab3 pattern, which indicate the stage-specific expression of Rab3. In adult males, Rab3 protein was found in the ejaculatory duct, subesophageal ganglion, and corpora allata. The highest concentration of Rab3 protein was observed in the subesophageal ganglion. The presence of Rab3 protein in the ejaculatory duct suggests that Rab3 may be involved in regulating sperm motility and/or acrosome reaction of the spermatozoon. In adult females, Rab3 was found in the corpus allatum and salivary glands. In addition to the sexual differences, variations in the Rab3 pattern among organs were also observed. The results of the present study suggest that Rab3, a small GTP-binding protein, has a key role in the transport of vesicles in Drosophila. with some others.
