By extrapolation, it is estimated that 2-8% of humans are infected with Mycobacterium tuberculosis (MTB) and 1.7 million people die of tuberculosis (TB) annually. However, the pathogenesis of the host response to this pathogen is not fully understood. The major focus in this field is the CD4+ T cell. Although memory T cells are critical for protection against pathogens, there is minimal information about how memory T cells can recognize and respond to a complex pathogen. Further, human studies on the correlation between cytokine production and protective immunity are lacking. Thus, information about how human T cells recognize and respond to M. tuberculosis (MTB) is lacking. My laboratory is developing the murine model of MTB infection, and we discovered that the mycobacterial antigen Ag85A can act as an agonist to stimulate Ag85B-restricted T cells in vaccinated and -unvaccinated mice. In addition, we discovered a non-specific T cell response to Bacillus Calmette-Guerin (BCG), which is the only live attenuated vaccine currently available to prevent TB. Based on these new findings, we hypothesize that there are differences in the T cell response to MTB in humans and mice that is important in determining the outcome of tuberculosis. Therefore, we will perform mechanistic studies to elucidate the requirements for the induction of a protective T cell response to MTB in the mouse model and determine if there are differences in the human system. In the first aim we will determine if the Th1 cytokines interleukin (IL)-2 and interferon (IFN) are required for protection against challenge with MTB; we will also compare the protective capacity of the mycobacterial strains, H37Rv and H37Ra, which differ only in their protein secretion profiles. In the second aim we will determine whether IL-6 is critical for controlling MTB infection; we will test this idea by comparing the protective capacity of IL-6 KO mice and IL-6 transgenic mice.
